At my biggest client we are setting up a local backup to a 3tb NAS and mirror to the amazon s3 cloud using zmanda. So far so good.

I use Crash Plan + to backup my desktop to the Crash plan cloud and have a Time Machine backup locally. Our laptops backup wirelessly to my MacPro via Crash Plan and they both have external drives that backup locally wherever they happen to be at the time.
Crash Plan + is awesome since you can pay monthly for cloud backup and you can use their software to backup laptops or other machines in your house to another machine at your home or business. Hell, you can even backup to a friends computer and visa versa.
